PCR & Agarose gel electrophoresis

Where your thermal cycle times the same as theirs? What about the voltage across the gel and the time of the run? Did you expose the RNA to light too much? It's possible you denatured them. Also, there may have been RNAses in your extract that could have reduced copy count drastically.

what is the meaning of Chlorophyll a/ chorophyll b ratio in

i don't know what the numerical value is for any particular organism, but it has to do with growing in low light. If the plant grows in very low light, it will accumulate more clorophyll b to capture more light, so the ration will decrease.
