Light-adapted eye

An eye that has been exposed to light, with bleaching of rhodopsin (visual purple) and insensitivity to low illumination.

Synonym: photopic eye.

PCR & Agarose gel electrophoresis

Where your thermal cycle times the same as theirs? What about the voltage across the gel and the time of the run? Did you expose the RNA to light too much? It's possible you denatured them. Also, there may have been RNAses in your extract that could have reduced copy count drastically.

what is the meaning of Chlorophyll a/ chorophyll b ratio in

i don't know what the numerical value is for any particular organism, but it has to do with growing in low light. If the plant grows in very low light, it will accumulate more clorophyll b to capture more light, so the ration will decrease.

Plants that grow under a green light will not died. Actually they grow a bit faster, BUT you will see a significant difference when comparing. The plant should come out weaker in some manner. Maybe the stem will be thinner or leaves might ...
