A protein involved in the regulation of cell adhesion, migration and spreading which serves as a negative regulator of integrin-mediated cell events and as adapter protein in the formation of the zone in osteoclasts.

Leupaxin gene is mapped on chromosome 11q12.1 containing 13 amino acids that belongs to the paxillin family possessed many protein binding domains implicated in targeting protein like focal adhesion kinase. It acts as transcriptional coactivators for androgen receptor and serum response that contributes to the regulation of cell adhesion that undergoes cytoplasmic shuttling containing nuclear export signals in its LD3 and LD4 motifs at the N-terminus while C-terminus contains four LIM domains.

Leupaxin is basically expressed in tissues and cells of hematopoietic origin such as in monocytes, osteoclasts and macrophages while it is highly expressed in lymphoid tissues like thymus, appendix, lymph node and vascular smooth muscle and its expression is vigorously regulated during development.

Leupaxin overexpression through transduction into osteoclasts evoked numerous cytoplasmic projections at the edge of the cell. Enhanced focal adhesion kinase activity attenuates nuclear buildup of leupaxin that limit the capacity to boost serum response factor dependent, thus it is relevant as a putative target for therapeutic interventions of prostate cancer cells.
