Competent cell growth on all my plates (multi resistant)

... given a batch of XL1 blue cells. I made them competent using calcium chloride procedure. To test them, i plated them out on agar plate containing: Kanamycin, Tetracyclon, Ampicilin an control. After overnight incubation, there is bacteria colonies in all plates. These colonies are so dense we cant ...

Re: Increasing Information For Genetic Algorithm

... of heterozygous individuals. Those genes can re-emerge when selective pressure is lifted. I will give you an example: You plate some bacteria on Kanamycin and wait. Most of the bacteria will die. However, a few days later you discover few very small colonies appear that are Kan resistant. If ...

Electrophoresis of a plasmid

... for an assignment was to predict to be the # of bands that would form on an agarose gel after electrophoresis of a plasmid which contained one kanamycin gene (1875 bp) and one ampicillin gene (3755 bp), and to explain our prediction. We didn't really learn much about this in class; it's more ...

Help Needed - Ligation Pairing (Vector and DNA)

... the reasoning behind these questions and how to understand the concept. Any advice, urls, would be appreciated. This experiment was cloning a kanamycin resistance gene into a plasmid and determining the orientation of insert. 1. Which of the following pairs could be ligated together? (All ...

No Band on agarose gel after Plasmid Isolation

... Plasmids it is working. But I can make a transformation with this approach and I get some colonies on my agar plate, which grow on my antibiotic (kanamycin). When I make a PCR from this plasmid isolation I see some bands, but no expected bands. The problem is that this is a mutated clone with ...
