Re: protein purification Ni-NTA... you see on the gel, or even remove them altogether if high or low ionic strength prevents binding completely. You can also try different IMAC resins to see if there are differences in the banding patterns, again suggesting that they could be non-specifically bound contaminants.

Ni-NTA-Columns... chelation matrix. USB, for one, sells a silica-based Ni resin they claim gives lower non-specific binding compared to either IMAC or agarose-type resins.

HELP: osteoclasts purification... (well, possibly not from G200, but even there I wouldn't think Sephadex could do much for resolving whole cells). There are modified Sephadex resins for ion exchange, but those would have names like DEAE-Sephadex, not Sephadex G10. So, I guess I'm skeptical that the method uses the G10 resin ...
