... could either plasmids with multiple insertion sites, make your construction in those (easy) then excise the double insert and transfer it to the genomic DNA as you would do. If you want to be more clever you can use a super PCR (I highly recommend Phusion Taq) to build your sequence and promoters ...

Re: Does complementary DNA include only coding sequences ?

Right - another technique, like RACE, is usually needed to extend the sequence across UTRs. If the primers are designed against genomic sequence, they can include UTR sequence, but are unlikely to capture the entire UTRs.

Aligning RNA to genomic DNA

... for gap open penalty and extension penalty setting - what I'm trying to do is to align an RNA seqeunce which is compose of exon 1 and 2 to a genomic DNA - however so far it I can only align either exon 1 or 2 but not both because there is a huge intronic gap in the gDNA. I tried reducing ...

molecular question.

... you have a mutant but you don't know the region where the mutant gene is allocated. How would you find out where the gene is located by using a genomic bank? I think it isn't possible with hybridization techniques because you dont have a primer? So im curious what the solution will be. Thanks :)

BL21 colony as a template

Hi.., I am working on E Coli metabolites. I want to amplify a 2kb product from BL21 cells. I do not have genomic DNA of BL21 so, I want to do colony PCR where colony as a template. Can anyone suggest me whether it is possible to amplify the product from colony PCR where colony ...
