RNA deletion

... the C and bump the codons, like this: AUG-UUG-AUA-CUU-AAU-UCU-GGU-C. See how the codons after the deleted nucleotide changed completely? This is a frameshift mutation. The codons will code for an entirely different amino acid. Hope that helps.

Introns and more

Darby is correct methinks.. But the process only works if the code causing the frameshift is found in an intron. ex. transcript with intron: |AAUUGG|AAUCGG|AAUUGG| mRNA: |AAUUGG|AAUUGG| - functional transcript without intron: |AAUUGGAAUCGGAAUUGG| mRNA: |AAUUGGAAU*********| ...

Re: Introns and more

Early-onset dementia? How does the presence of an intron downstream from a frameshift in genomic DNA undo the frameshift in the mRNA?

Re: splicing MO vs translation MO

... a splice modification will eliminate protein activity, we usually aim at either skipping the exon that encodes the active site or triggering a frameshift that, by introducing a premature stop, triggers nonsense-mediated decay of the transcript. As for your splicing Morpholino, how much larger ...

protein sequence

... the final G. It would have been nice if they had given you more of the sequence to the right of what we see so we can tell if there really is a frameshift or not. You can’t even say what the ninth residue of the mutant protein is without that information. My answer, to be honest, is that there ...
