Dictionary » F » Fab FabFab (Science: immunology) fragment of immunoglobulin prepared by papain treatment. Fab fragments (45 kD) consist of one light chain linked through a disulphide bond to a portion of the heavy chain and contain one antigen binding Site. They can be considered as univalent antibodies. ![]()

Results from our forumProtein bonding(antigens)... is not covalent. It has to do with chemical noncovalent bonding/binding. Here is a picture that shows the "pocket" of the antibody Fab fragment with an antigen (epitope): http://www.pnas.org/content/102/14/4984/F3.large.jpg and a space filling model: http://www.google.com/imgres?imgurl=http://www.scripps.edu/newsandviews/e_20050228/enlarge1.jpg&imgrefurl=http://www.scripps.edu/newsandviews/e_20050228/enlarge.html&usg=__zru4TKFYtwhCQl1ThS6a-gxA5ZY=&h=403&w=324&sz=26&hl=en&start=105&sig2=eAIM5QTonqwSAsvCSkBYJQ&um=1&itbs=1&tbnid=26BBr_-5Z4oDPM:&tbnh=124&tbnw=100&prev=/images%3Fq%3Dantigen%2Bbinding%26start%3D84%26um%3D1%26hl%3Den%26safe%3Dactive%26sa%3DN%26rlz%3D1T4GGLL_enUS309US312%26ndsp%3D21%26tbs%3Disch:1&ei=YwP8S-qFKZvsNcC3jDk

Fab library constructing-ligation/transformation problemI am working on constructing a phage library to express antibodies and currently I am doing the Fab library constructing work. I did PCR and got light chain (LC) products. There is a LC stuffer in the vector so I digested and gel purified the digested phagemid vector and ...
