How to prepare a 3.5% agarose gel ?

I cooked it on a hot plate on low temp with a stirring magnet and in the end put it in a microwave oven on low-medium for a couple of soconds a few times to get rid of the bubbles. and after getting a clear solution i added the atidium bromide. worked great.

MOLECULAR CLONING

... I understand, it is not necessary to dephosphorylate my double-digested open vector as long as it was cut with two different enzymes. Thus, the 5'-end of my vector has a phosphate. But still, should I phosphorylate my insert, since its 5'-end does not have a phosphate...?
