Empty

1. Containing nothing; not holding or having anything within; void of contents or appropriate contents; not filled; said of an inclosure, as a box, room, house, etc.; as, an empty chest, room, purse, or pitcher; an empty stomach; empty shackles.

empty is used as the first element in a compound; as, empty-handed, having nothing in the hands, destitute; empty-headed, having few ideas; empty-hearted, destitute of feeling.

Protein ladder width increases down the gel?

... protein that I loaded separately and combined in the ladder are from the same solution. In addition, I loaded 1X loading buffer dye in the empty wells. Any ideas? Also, I'm afraid that the bands will not be as prominent if I dilute it further. Thank you in advance!

MOLECULAR CLONING

The purpose of dephosphorylation is to prevent self-ligation of empty vector. So unless your insert is very long, it should not be necessary.
