Dnase i

a type of endonuclease, or enzyme that breaks down dna molecules at their phosphodiester bonds. It is found in large quantities in the pancreas.

Have you even checked the gram stain?! If you did, I may be able to tell you off the top of my head. How can you be working on identification and not have a manual? It doesn't have to be the one you mentioned, other people have published manuals too. Try a library.

Of these four bacteria are any positive for for DNase? I've searched and searched and cannot find (reliable) information on this. I do not have access to Bergey's manual. Pseudomonas aeruginosa, Pseudomonas fluorescens, Proteus mirabilis, and Citrobacter ...

... I guess the only tricky part will be the preparation of DNA. To clone it, I will need manageable sizes of DNA so I might have to treat it with DNAse or nucleases. How much should I add? For the electrophoretic mobility shift assay, how would I detect it? Maybe a Western blot?

Put the gene under the control of an inducible promoter. That way your introduced DNAse can be selectively expressed.
