Re: protein purification Ni-NTA... if high or low ionic strength prevents binding completely. You can also try different IMAC resins to see if there are differences in the banding patterns, again suggesting that they could be non-specifically bound contaminants.

protein interaction... it was because my proteins are secreted protein, we know for 2-hybrid, the reporter genes can only be motivated when the two proteins of interest banding to the AD and BD separately enter into the nuclear. so maybe the 2-hybrid has some limits I think. Well...... I used EcoRI and HindIII to verify the insert based on the size of bands generated. I.E. I am fairly certain the insert is present based on the banding pattern generated from cutting the pBS-insert with EcoRI and HindIII.
