Results from our forumseparation problem with GelRedHello! We recently switched from ethidium bromide to Gelred in our lab. (We always prestain gels - poststaining is no option because its too inconvenient and timeconsuming - using 1xTAE buffer). Our DNA samples and the ladders aren't separated ...

Calculating DNA concentration in PCR product... to dsDNA. It has a linear range from at least 50-800 ng DNA/mL. With special assays you can go down to 5 ng/mL. Other options would be Ethidium bromide (which is around 10 times less sensitive) or the fluorescent dye SybrGreen.

Why does DNA fluoresce under UV light?... that it is the DNA in body fluids that make them fluoresce under UV light but I'm not sure why. If that's true, why do we need to add ethidium bromide to visualize DNA gels, wouldn't the DNA be visible by itself? Is it just a mater of intensity? I also heard that body fluids have to be dry ...

Does my protein bind DNA?... also release DNA first and run it on a regular TBE-agarose gel. If sufficient amounts of DNA present, you will be able to visualize with ethidium bromide. Western is another possibility.

SYBR gold in acrylamide gelYou have anything in your gel or buffer system that quenches fluorescence? Ethidium bromide, maybe? I don't run SYBR gels, so I don't know if that sort of thing happens or not. Another possibilty, I suppose, is that your sample got stuck in the well, but if there ...
