coomassie staining

hi
I will run some protein gels today. Now I have one question: I will stain them with coomassie. Is it possible to destain them over night? or will the protein bands dissappear ?? :?

You should be OK overnight in the standard destain of 30% MeOH, 10% HOAc. It is possible to completely destain a coomassie-stained gel, but you usually up the MeOH to 40-50% and go for at least 24 hours. You can leave the gel in stain overnight and destain in the morning, or transfer a partially destained gel to water for the overnight, and continue with destain in the morning. All of that works.

U wash the gel with 45-50 C hot water.it'll really work.gv time for wash abt 20-30min each.After 4-5 wash it'll destain easily.[/code]
