Poly acrilamide gel electrophoresis (P.A.G.E.)

Postby unlucky jester on Mon Jan 17, 2005 11:11 am

I have to list ten possible sources of procedural errors which may affect the accuracy of P.A.G.E.
Can anybody help?

we have lots of trouble shooting with PAGE.

List them.... I'll try...

I haven't resreached this but I'll give it a try:

accuracy of measuring equipment such as pipettes,

how similar the background colour of gel might be to those of the bands (I had that prob in a prac),

the logarithmic measurements of the distances the bands travel, ie - the nearer these are to the beginning the closer their size values are so a band could be measured as lets say 7 kb when it's really 15 kb (that happened to me in another prac),

not sure but would amount of electric current and how long it was on affect how far the bands run? I would think it would possibly.

That's all I can think of for now.

do you know how the principles of gel elecrophoesis allow for DNA seperation?

It'll be many books, a good one is principles of biochemistry but it's in tounes of others. I could explain it but just look it up and then if there's something specific you can't find ask here. Why do so many members not look in books, just post their questions straight here? :?

Hi Unlucky Jester,
I am sure that that the passage of electric current would be a very important factor in PAGE. The problem arising due to insufficient contrast between the band color and gel background seems quite unlikely. Not sure...But I am sure of the electric current factor.
