How to detect long genomic DNA after extraction

Postby daniell on Thu Feb 23, 2006 11:10 am

My genomic DNA is around 6.1Mb. After DNA extraction, how am I going to confirm that I have extract the DNA?

you can measure absorbance of light at 280 nm and 260 nm The absorptionmaximum of DNA/RNA is at 260 nm and of protein at 280 nm

--> when pure DNA is isolated, then the ratio 260/280 will be approximately 1,8

Now you do know there is DNA (or not) and how pure it is.

Another way is a restriction digest using restriction enzymes and running the samples afterwards through electrophoresis. I do that all the time at uni. Info on electrophoresis can be found in tounes of bio textbooks.
