seperate nucleotides by electrophoresiswhat is the best pH value when seperating AMP UMP CMP GMP in electrophoresis?
Thanks for your attention!
use a slightly acidic pHto separate those nucleotides. Try using Tris EDTA buffer adjusted to pH = 6.25 as the mobile phase on your agarose gel.
Behold God's handiwork--it is in the biological universe and in your Mind. Praise Him!
"Nucleic acids however, remain negative at any pH used for electrophoresis and in addition carry a fixed negative charge per unit length of molecule, provided by the PO4 group of each nucleotide of the the nucleic acid. Electrophoretic separation of nucleic acids therefore is strictly according to size."
so, my opinon is just give a PH which can not destroy these nucleotides
Thanks!pH A, 1.5 B, 3.5
C, 6.5 D, 9.5 I don't know why and which one I should choose .
