low levels of protein but high activity levels

Hi,
I am stumped on one of my projects. In my experiment I found that the gene expression is high, protein levels low but activity levels are high for this enzyme in diseased samples compared to age-matched controls. Has anyone else seen anything like this?

Now seriously. Do you know what is the specific activity of your pure enzyme? That could tell you something. How do you determine the protein level?
