E.coli GenotypeHi,
I am trying to generate a minimal media for the E.coli strain with the following genotype F'[lacIq Tn10(tetR)] mcrA Δ(mrr-hsdRMS-mcrBC) φ80lacZΔM15 ΔlacX74 recA1 araD139 Δ(ara-leu)7697 galU galK rpsL(StrR) endA1 nupG with the addition of Leucine I have managed to grow them, but the yield is anything but great. I am a little confused about the combination of araD139 and Δ(ara-leu) in the genotype. araD139 means, that a mutation in L-ribulose-phosphate 4-epimerase blocks arabinose metabolism and Δ(ara-leu) means, that the ability to built arabinose (and leucine) is deleted. Can somebody help me and tell me if or if not my E.colis need arabinose? And/or if they are missing something else, please? Thank you Kiki
Re: E.coli GenotypeI believe that the ∆(ara, leu) simply means that your strain of bacteria cannot catabolize arabinose or leucine.

I am using TOP10 and it has the same deletion. My cells do happily grow on M9 or the minimal medium of our group with leucine. DH5a doesn't have it I think.
