can i confidently detect an e.coli in water just by performing the indole test alone?

No - you'd have to develop a test for indole in mixed culture of direct water inoculation and other bacteria produce indole (from tryptophane) including Vibrio, Pleisomonas, Salmonella, Citrobacter spp.

The indole test was done ofcourse after the presumptive to complete the test, so what i should do is perform further biochemical test other than indole to identify an e.coli. Is API 20E more realible than IMVIC test?

Presumptive? Sorry, I thought you were suggesting unique protocol to identify directly presence of E. coli in water. What is the purpose then? If a one-time experiment, use API.

ah ok, its not practical to use API always,,, its more expensive than the IMVIC but API can give rapid results..thnks sir

You could do it the old way - smell, colony morphology on EMB and Mac
