plasmid cut and uncutHi there,
I have to bands on my gel..one uncut plasmid and one cut...they were more or less the same size with the cut being slightlyyy bigger..(looking at them both compared to the ladder)....Just wondering if there is a reason for this?
Re: plasmid cut and uncutYes, plasmids show different mobility depending of their conformation. the fastest= supercoiled. above this linear digested and above these relaxed nicked circular form check any manual to confirm. Buena suerte
the uncut plasmid should have more than one bands. I have seen usually two, but I have read about up to 5. Onthe other hand the cut vector should show sharp single band.

Cis or trans? That's what matters.
