Freezing Het-1A cellsModerator: BioTeam

Freezing Het-1A cellsI recently obtain via ATCC Het-1A cells. Their product info sheet says that I must use the following cryoprotectant medium: L-15 plus 10% FBS, 1% PVP, and 7.5% DMSO. I did order the specific complete growth medium BEGM kit but I want to know if I can just DMSO to this growth medium in order to freeze or if it is necessary to get the cryoprotectant medium?
in my tissue culture days i used to just add DMSO to whatever media I normally grew the cell lines, and it worked for the lines I worked with. However, i never worked with Het-1A, it may be that this particular line is more sensitive

Re: Freezing Het-1A cellsHi! I don't know if this is going to be any help at this stage but I work with this cell line and I freeze them in 90%FBS and 10% DMSO. This works really well for me as long as you remove, by centrifugation, the above mentioned mixture when you defrost the.
I have now been using this method for 3 years, I did try to use a commercially available freezing solution but it actually reduced my cell viability when reviving cultures. Hope this helps someone out there : )
