Spore counting

Postby pinkypig on Mon May 05, 2008 11:57 am

What is the procedure for visualising spores to perform a total count on them? Taking into account that spores are difficult to stain.

For fungus I know they stain with calcofluor white and then count with epifluorescence microscope.
Looking at he litterature other methods: are:
-Heat shock to reactivate the spore and plating and suitable medium
-Haemocytometer, but I guess that must be for eukariotyc parasites
