jelly fish protain

hi every body. how are you
there is a protain in the jelly fish that emits light (GFP). if i extract this protain from the cells. how can i make it emit light? i guess what im asking is how can that protain emit light. what chemicals must be present?

The protein only floresces green when hit by blue light, it does not generate light. Squid do not have it.
You might wanna look into luciferase. Luciferase oxidises a substrate and converts the released energy into light. Unlike GFP, luciferase actually "produces" the light

the luciferase enzyme has been "discovered" by bacteria, fungi, sea anemones, dinoflagelates and fireflies. Here is a link to a page describing luciferase. Very cool. I have actually printed all those files and made a molecule catalog

The luxCDABE operon can be isolated directly from Photorhabdus luminescens DNA. It gives luminescence with an enzymatic reaction (needs O2 and is Temperature dependant).

As for GFP you just need the right bulb to make it fluoresce.

I did not have to search because in one of my projects I have to work with a plasmid containing the lux operon.

@Medhi:
Don't ask, I cannot give you the strain, it come from another lab and we cannot share the strains, they have not published the construct. But it works.
If you can find it somewhere else? I would do a pubmed search with lux and E. coli as keywords and then contact researcher that have worked with such constructs if they exist.
