In step 3 of the Krebs Cylce, Isocitrate undergoes oxidative decarboxylation to produce a-ketoglutarate (CO2 and NADH by-products). This is catalyzed by the Isocitrate dehydrogenase enzyme.The reaction involves dehydrogenation to Oxalosuccinate, an unstable intermediate which spontaneously decarboxylates to give a-Ketoglutarate.

Then in step 4 the a-ketoglutarate undergoes oxidative decarboxylation to produce Succinyl-CoA (CoA-SH, CO2, and NADH by-products). This step is performed by a multi-enzyme complex, the a-Ketoglutarate Dehydrogenation Complex. The multi-step reaction performed by the a-Ketoglutarate Dehydration Complex is analogous to the Pyruvate Dehydrogenase Complex, i.e. an a-keto acid undergoes oxidative decarboxylation with formation of an acyl-CoA.
