How do you work out DNA concentration?

I have spec DNA- result 0.023
Dilution 1/1000

Should my answer be 1150ug/ml?

How do i translate this to ng/ul?

Hi angela.morris

You are right your concentration is 1150ug/ml

What you read with spectrophotometer is "Absorbance" of your sample
you have to multiply this number for a coeffiicient that is 50 for DNA and for your diluition factor (1000 in your case)
0.023 x 50 x 1000= 1150ug/ml

(This formula is known as Lambert-Beer)

To convert into ng/ul is very simple: is exactly the same 1150ng/ul

Sounds correct. Just to add that the conversion to ng/ul from ug/ml is exactly the same because ng is 10-9, ug 10-6 and ml 10-3. Thats 10 to the power of -9 etc. Therefore 10-6/10-3 is same ratio as 10-9/10-6. Works if you do the maths.
Please correct me if im wrong.

I used it. In my 1st and 2nd year of university. Everyone who was doing a biology related degree at the uni had to use it for the basic genetics and molecular biology practicals.

Im new here. I've never mentioned anything about university before. :?
Im in my 3rd year of a 4 year Cell Biology degree. So ive only got just over a year left :shock: :o
