Designing Primer for over lapping PCR

Postby Rezaulbiox » Tue Feb 28, 2012 7:33 am

Hello everyone,
I am trying to amplify a little bit long DNA of 6.72kb. I designed primer within 100 bp upstream and downstream of this DNA but failed to get the DNA by PCR. Some one suggested me to amplify the DNA by overlapping PCR. I do not know how to design the primer for overlapping PCR. I usually used Primer Premier 6 as primer designing software.

You are highly appreciated to give me some guidelines to design overlapping primer by using Primer Premier 6.

Thanking you
