Question about Kirby-Bauer method

Hi, I'm trying to do the Kirby-Bauer method by testing the effect of certain foods' ability to inhibit pathogen growth.

I was wondering if it's possible to do this method by just streaking the agar plates with pathogens like E.Coli straight from the tube (from the catalogue) or is it a must to use only pathogens after it has been grown in broth??

My plan was to just streak like E. Coli with an inoculating loop in a zig-zag pattern every 90 degrees, pretty much covering the whole plate. And then I would place my paper disks on top of the agar...

Usually you do overnight culture first. First, you will make sure the bacteria are viable. Second, you don't want to run out of your stock and order it again
