yeast expression vectorHi All,
I’m looking for a vector for expression of a third bridging protein in the Invitrogen Proquest yeast two-hybrid system. Does anyone have or know of a yeast expression vector that suits the following requirements: • High expression promoter that is independent of GAL system. • Marker for selection of transformants in yeast that is not Leu, Trp, His or Ura. I currently have clones set up with the Invitrogen two-hybrid system to test specific protein interactions, but I’ve found no interactions and recent publications have shown a third protein acts as a bridging protein for homologs in other species, so I’d like to try adding this protein in to my existing clones. The yeast strain is S. cerevisiae Mav203 [MATα, leu2-3,112, trp1-901, his3Δ200, ade2-101, gal4Δ, gal80Δ, SPAL10::URA3, GAL1::lacZ, HIS3UAS GAL1::HIS3@LYS2, can1R, cyh2R].
