please i would know th protocol of AS-pcr and what is th difference between it and ASO-pcr.

As far as I am aware AS-pcr and ASO-pcr are the same technique and use allele specific oligonuleotides (primers) in SNP detection.
There are plenty of protocols available online and on Springerlink for allele specific PCR.
