Development of a hybrid delta-endotoxin and its expression in tobacco and cotton for control of a polyphagous pest Spodoptera litura

Abstract A hybrid -endotoxin protein was designed against a polyphagous lepidopteran insect pest Spodoptera litura, which is tolerant to most of the known -endotoxins. The hybrid -endotoxin was created by replacing amino acid residues 530–587 in a poorly active natural Cry1Ea protein, with a highly homologous 70 amino acid region of Cry1Ca in domain III. The truncated -endotoxins Cry1Ea, Cry1Ca and the hybrid protein Cry1EC accumulated in Escherichia coli to form inclusion bodies. The solubilised Cry1EC made from E. coli was 4- fold more toxic to the larvae of S. litura than Cry1Ca, the best known -endotoxin against Spodoptera sp. None of the two truncated toxins, solubilised from E. coli caused larval mortality. However, trypsinised Cry1Ca protoxin obtained from E. coli and solubilised from inclusion bodies caused mortality of S. litura with LC50 513 ng/ml semi synthetic diet. A synthetic gene coding for the hybrid$-endotoxin Cry1EC was designed for high level expression in plants, taking into consideration several features found in the highly expressed plant genes. Transgenic, single copy plants of tobacco as well as cotton were developed. The selected lines expressed Cry1EC at 0.1–0.7% of soluble leaf protein. Such plants were completely resistant to S. litura and caused 100% mortality in all stages of larval development. Hence, unlike in E. coli, the hybrid -endotoxin folded into a functionally active conformation in both tobacco and cotton leaves. The truncated Cry1EC expressed in tobacco leaves was about 8-fold more toxic (LC50 58 ng/ml diet) compared to expression in E. coli.

I have worked 5 years with the people that published those results, and indeed handled and worked on the collection described in this paper. Believe me, although the name may seem frightening, those mutators are not going to killl us before long. I could suggest you to read about R. Lenski's experiments and you would see that it takes more than 10000 generations to start obtaining a bacteria close to become a new species.

I suggest you to start reading a lot more about introductory biology before worrying bout snippets that you do not seem to fully understand. Because when I see extract of a book on how to have a good post-doc, I really wonder what you are posting. This guide is probably useful for the like of me in the US, but it is nowhere near to be a guide on how to rule the world (I wish I simply had more money than a freshly hired technician...) or to create any deadly disease. And believe me post-docs work hard and are full of good intentions (sometimes a bit selfish, like to get a real job), and I stilla have to meet any scientist likely to even consider creating pathogenic agents for the pleasure...

This topic has been overlooked and not discussed for far too long.

How to spot a dis-information specialist.

All who are infected with this illness and have been watching all the discussions concerning it are aware of some items. These I will discuss here today. First of all there are many interest groups set up to defend this issue. They have done so primarily by enforcing/ promoting the delusions of parasites theory. We know this as fact but have you considered exactly how they have and continue to promote this?

First of all, many of the people who are active in this discussion do not have this illness(though many say they do). They are paid by various interest groups to discredit the illness and also those whom have it. It is their job to monitor the different discussion groups and sabotage any real organization or logical posts made by anyone. this happens with a number of controversial topics, including this one. they are not unique to this illnesses discussions. That said lets look at some of the ways they do this.

Ways to spot this type of behaviour
1. Use derogatory language and fly off the handle.

2. they will call others names and make insinuating remarks and otherwise say anything to discredit any valid points being made concerning the topic. they will question others character often.

3. They will most often have the largest number of posts of any of the sites other members. Often by 10 to 20 times as many posts as others making valid points in each particular discussion.

4. Often they will be the very first person to reply after any thought provoking post concerning the topic is made. NOTE: they will often throw out a string of unrelated information and links pointing in many different directions to confuse the point previously made.

5. they present an overabundance of information and avenues of direction to follow up on for those reading/researching. Most information upon looking further into actually has NOTHING to do with what is being discussed. This also is meant to confuse an issue and also serves to make all involved to appear mental or conspiratorial. THIS ALSO SERVES IN MAKING OTHERS LOSE INTEREST IN TOPIC simply because of extremely long discussion threads with an enourmous amount of unrelated information. In our case concerning this illness, I have been told by a dozen or more people that some threads are avoided simply by seeing particular people having been an active participant in the discussion. They annoy people to the point of losing interest, no matter how interesting a topic may be.


6. They claim to have the answer constantly, but each time the answer is different from the any other previously made. Look for the phrases “This is it” or “like I have said before” “I know this is involved” esp. when topic is known to still be a mystery. They will say anything to confuse the reader and get them off track. they are extremely non consistent in thier beliefs.

7. they are non focused and each day will present new and “important information” which is most often non related to their other important info posted previously.

8. often remarks made by them out of frustration is then censored and deleted from discussion.

9. they are intelligent people, but because of the constant posting and monitoring of the discussions, appear to be unemployed. Note: simply tract the times of which their posts are made. Most people will post around the same time of day because of work, family and other responsibilities and obligations.


Ok I think I have made my point. This is not a complete list of methods involved by these people, not by a long shot. But you hopefully get the point and can now start to see through the smoke screen they are responsible for. Do not doubt these people exist. They do and are very active in all types of debatable topics. Trust your gut feelings as they are most often quite correct.

confusing. I'm glad that you wrote and wish you all would do so more.

Let me ask you a question. Do you see Bt. as being the same thing as
anthrax? I do and it seems many scientist will agree with that as well, but I would like to hear what you think about that.

Effect of electrical charges and fields on injury and viability of airborne bacteria.

In this study, the effects of the electric charges and fields on the viability of airborne microorganisms were investigated. The electric charges of different magnitude and polarity were imparted on airborne microbial cells by a means of induction charging. The airborne microorganisms carrying different electric charge levels were then extracted by an electric mobility analyzer and collected using a microbial sampler. It was found that the viability of Pseudomonas fluorescens bacteria, used as a model for sensitive bacteria, carrying a net charge from 4100 negative to 30 positive elementary charges ranged between 40% and 60%; the viability of the cells carrying >2700 positive charges was below 1.5%. In contrast, the viability of the stress-resistant spores of Bacillus subtilis var. niger (used as simulant of anthrax-causing Bacillus anthracis spores when testing bioaerosol sensors in various studies), was not affected by the amount of electric charges on the spores. Because bacterial cells depend on their membrane potential for basic metabolic activities, drastic changes occurring in the membrane potential during aerosolization and the local electric fields induced by the imposed charges appeared to affect the sensitive cells' viability. These findings facilitate applications of electric charging for environmental control purposes involving sterilization of bacterial cells by imposing high electric charges on them. The findings from this study can also be used in the development of new bioaerosol sampling methods based on electrostatic principles. Copyright 2002 Wiley Periodicals, Inc.

Please do not use the deaths of these poor souls as a means to disprove or disrespect others who are actually trying to help the cause and prevent more of that very thing.

You speak of openness and truthfulness.
Putting all ones private info out for all to see may be fearless ...... but truthfull it does not make. No one is obligated to divulge personal info and they are being smart by not doing so. Why continue to entice people to do this?

First of all my site is not filed with untruths. If so, straighten me out and I will correct them with facts. Give me some solid facts and I will change my site..but I guess you will nto do that either..am I correct?

Second..I am not using anyones death for my own advantage. In fact I am doing some good between two people...and am sensative to the reasons why they did what they did....... so you really need to know what you are talking about where that is concerned. False hope kills.

Third, I never forced anyone to give out anything. But I do and will continue to since I know that people need a phone to phone support group and not just one with faceless, voice-less individuals who you have no idea who they really are or where they come from or what they are really up to.
