Creating Cell Lines

I'm trying to figure out how you would create a cell line that is missing certain receptors. Would you ablate receptors, or are there too many receptors for that? My other two ideas would be that you would somehow permanently bind something to a receptor or somehow silence the receptor in the genome.

If you have a protocol on how it's done I would really appreciate that. Thanks!

Is the receptor in GPCR/Kinase/any other enzyme family? If you know the receptor, you should try to knock it out. You need a stable cell line though. That is a big project. But many times, you get multiple receptors for one ligand. siRNA could be used for silencing.
