Conservation of samples/DNA during an expeditionHi!
I am preparing my first expedition. I will be going to Africa to study mycorrhizae. To do so, I will be taking cores around trees. I probably wont have much of a lab available, and it will be several weeks before I am back to England or the USA. What would you advise? I was told that I could maybe sort my roots and fungi on site and use CTAB buffer, but the method seems quite complicated and apparently requires a lot of sample material, so I am not sure if it can be applied to microscopic roots... Any idea is welcome! Thanks a lot
You could probably resuspend your samples in tubes containing a reagent that would denature proteins like Guanidium isothiocyanate (GITC) or Guanidium Hydrochloride and freeze/refrigerate yoour samples until you go back to your lab.
