Lookin for a genomic DNA extraction kit for bacteria

Postby genetim » Fri Mar 07, 2008 3:53 am

Hi all,

I'm looking for a good genomic DNA extraction kit for bacteria. Do you guys have any suggestion?
By the way, what criterion do you have to choose a suitable kit?

Your great help is appreciated.

By the way, do you use kits or home-made method for bacterial genomic DNA extraction? Why?

Thank you very much.

The Qiagen Sidewinder Genomic DNA Purification kit works well for me. I never make my own reagents any more if I can buy a suitable kit, although I will occasionally make up extra buffer when needed.

I use the mobio kits because I work with environmental samples and they consistantly extract all kind of DNA correctly. But it depends what application you have for your DNA. Many kit do creat a lot of shearing in your DNA, and that is not always a Good Thing.

Sorry. Novagen makes the Sidewinder kit, though I bet Qiagen has an equivalent one. I'm not familiar with Mobio, but I suspect it works just as well. These kits all tend to be good.

They are all based on the same technology, but Mobio is really good for environmental work, because it is in my experience the best at removing PCR inhibitors from samples. And besides it uses a bead beating lysis, instead of enzymatic one. And that solves may problems, besides being faster. But it shears the DNA a lot and it is slightly more expensive, so it really depends on your applications.

Brand MoBio looks great. For me, time is an important parameter, both PCR and sequencing are required in downstream. I wish this kit may help.

Thank you for suggestions.
