One question about Fluorescence in situ Hybridization(FISH)

All protocal suggest that Fluorescence in situ Hybridization(FISH) need humannity chamber. Is this instrument necessary? and why? I want to use BAC clone as probe (labelled with DIG-Nick Translation MIX, from Roche) and detected with Fluorescent antibody enhancer set for DIG Detection (also from Roche) .
