transient transfection with luciferase system

Hi,

I do now transient transfection with the promega luciferase sysyem (FireFly and Renilla Luciferase) and would like to know how long the constructs are in the cells. Because I learnt that 2 days after transfection the cells "skip" the plasmid but I would do longer treatments and would like to see if there is a time dependency. But if the cells loose the plasmid before I measure the luciferase I'm not able to see any effect...
Hm, maybe someone has a few experience with this topic or is very smart and can help me?!

hello.
In our lab we are doing Luciferase assay....
one option is to do double transfection of your plasmid....
for example: firstly you do transfection and next day after that or two days after first trasfnection you can do transfection again...
