cellulolytic nitrogen fixing bacteria

i wanna know how to screen the cellulolytic activity of this bacteria. which is the best method? and which method can i use to measure the nitrogen fixing activity? i want to achieve overproduction of nitrogen from the broth culture of these bacteria. please, help me.

Gives details of celluase activity determination by standard filter paper assay (essenetially measuring the enzymatic release of reducing sugars from paper).

Nitrogen fixation is not producing N2. It is taking N2 gas out of the atmosphere and converting (i.e. fixing) it to NH3 (or its equivalent). You could measure the rate of N2 consumtion manometrically, I suppose. A proxy for measuring nitrogenase activity is to use acetylene reduction, but you’ll have to look up a method. I don’t know the techniques well enough.

I’m not sure what to suggest for trying to optimize growth conditions.
